Lipophilicity is a primary physicochemical parameter studied in early drug discovery. LogD data can aid in the understanding and prediction of passive permeability, metabolism, excretion, oral bioavailability and toxicity. It is strongly correlated to solubility and characterizes the volume of a molecule in a specific environment.
Building upon widely recognized chromatographic principles, we have developed AlphαLogD, a high-throughput assay optimized to determine LogD at pH 7.4. Initially developed to measure small molecules, we have found that this assay extends to larger molecules in the beyond-rule-of-5 (BRo5) chemical space. The graph below contains data from an internal validation study. A calibration curve was built using the average of 5 measurements for 25 commercially available compounds.
